Overview

Accelerated Biosciences (AB) human Trophoblast Stem Cells (hTSC) were obtained and cultured under current Good Tissue Practice (cGTP) and conform to Global EthicalStandards and Clinical Cell Regulations.
 
All AB hTSC cells are manufactured by AB’s CDMO partner based on AB’s standard operating procedures.  ABX-002 cells are free of pathogens such as mycoplasma, bacteria, yeast, and fungus, and are guaranteed to achieve > 70% viability post-thaw at the indicated cell number.

ABX-002 cells are intended for Research Use Only (RUO) and shall not be used for clinical or diagnostic purposes. Corresponding GMP cells are subject to commercial license.

Unpacking & Storage
  • Check all containers for leakage or breakage to ensure integrity
  • Remove frozen cells from the dry ice packaging or cryogenic container and immediately transfer cells to storage below -150°C until use
  • Store hTSC in vapor phase liquid nitrogen (LN2)
  • Avoid temperature excursions to prevent product degradation
Thaw
  1. Remove hTSC from storage and maintain temperature until final thaw
  2. Thaw hTSC quickly in a 37°C water bath or mechanical thawing device
  • When using a water bath, swirl the sample gently until only a small ice crystal is visible
  • The approximate thaw time for a 1 ml sample in cryovial format is 2-3 minutes
  • The cryovial should be cool to the touch and SHOULD NOT warm to above-chilled temperatures
Recommended Culture Procedures
  1. Once thawed, the cell/cryomedia mix should be diluted into culture media (e.g. MesenCultTM - ACF Plus) at a ratio of 1:10 sample to media
  2. Remove a sample to determine cell viability and number
  3. Plate hTSC into an appropriate culture vessel (recommended density below) and allow cells to adhere for at least 4 hours; incubator atmosphere: 37°C, 5% CO2
  4. Once cells have adhered to the culture vessel, media should be exchanged with fresh media for downstream culture or differentiation
Recommended 2D Culture Procedures
  1. Plate hTSC at a density of 6,000 cells/cm2 in appropriate culture media; we recommend MesenCultTM - ACF Plus
  2. Sub-culture the cells every 3 to 4 days at the following specified range of seeding densities; during this time frame, no medium change is necessary
  3. Seed cells at a density of 5,000 – 7,000 cells/cm2 if cells are passaged after 3 days or 4,000 – 6,000 cells/cm2 if cells will be passaged after 4 days; refer to the following table for the recommended seeding cell number and volume of culture medium
  1. Note: Cells should never be allowed to reach a degree of confluency greater than 70 - 80% (~ 40,000 cells/cm2) as contact inhibition decreases the number of population doublings the cells can undergo before reaching cellular senescence.
